The milestone match saw the Nigerian duo of Victor Boniface and Nathan Tella play a part in the occasion, with the former watching from the stands due to injury and Tella coming on as a second half substitute.
Leverkusen broke the deadlock through a fine Xhaka strike, with the Swiss midfielder celebrating with a cheeky fake hamstring injury. The visitors fought back and equalised five minutes after going behind through a loverly header from Dominik Kohr.
Mainz goalkeeper Robin Zentner then pushed Robert Andrich's shot into his net to make it 2- and give Alonso and his brilliant Leverkusen another excellent night at the BayArena.
By usurping Bayern Munich's record unbeaten streak, Die Werkself have underlined their status as Germany's new dominant force under Alonso.
With an 11-point lead and game in hand, Leverkusen are firmly on course for their first Bundesliga title since 1993.
