Otieno, who played as a left winger, provided an assist with his first touch of the ball when he received the ball down the left flank before whipping in an inswinger towards Chris Popov, who headed it home to give them the lead less than two minutes played.
Otieno has been in good form for Leicester when fit. The former Chelsea star has scored five league goals and made four assists in 16 PL2 appearances this season.
Leicester City's dominance was further highlighted by goals from Sammy Braybrooke and Will Alves within the first 33 minutes of the match, as they ran riot in east London.
Although West Ham's Kamarai Swyer managed to pull one back, overturning three Leicester City goals in the first half proved to be an insurmountable task for the U21s.
