Farouk Tumwesigye doubled the lead, again three minutes into the half, to express their intent.
Ibra Ssebagala added the third ten minutes later before Frank Sekanjaako and Leonard Kasenyi stretched the lead.
Nkoola, who had opened the scores, sealed the result in the 76th minute.
The result takes Vipers top of the five-team table with seven points after two games.
A win with a margin of more than two goals guarantees a team an extra point.
BUL FC JT are the defending champions after winning the last edition n 2019 while Vipers are two-time winners.
